Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp's are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Grp94 has been studied as a prognostic factor in breast cancer. Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa. This MAb recognizes an intermediate filament protein of 46 kDa, identified as cytokeratin 20 (KRT20). KRT20 is abundantly expressed in goblet cells and enterocytes of the gastrointestinal tract. It is a useful marker of pancreatic and colorectal cancer. KRT20 is expressed under normal, hyperplastic and neoplastic conditions. It has been detected in adenocarcinomas of the colon, stomachand biliary tract. Breast carcinomas are generally non-reactive. Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In non-muscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a muscle-specific, alpha actinin isoform that is expressed in both skeletal and cardiac muscles. The binding epitope of this antibody is located in the first EGF-like repeat domain (EGF1) between amino acids 27-59 of Ep-CAM. EGP40 is a 40-43 kDa transmembrane epithelial glycoprotein, also identified as epithelial specific antigen (ESA), or epithelial cellular adhesion molecule (Ep-CAM). It is expressed on baso-lateral cell surface in most simple epithelia and a vast majority of carcinomas with the exception of adult squamous epithelium, hepatocytes and gastric epithelial cells. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. Recognizes a protein of 31 kDa, which is identified as MTAP (5'-deoxy-5'-methylthioadenosine phosphorylase). It catalyzes the reversible phosphorolysis of methylthioadenosine, which is important in polyamine metabolism and for the salvage of adenine and methionine. The gene encoding MTAP is linked to the tumor suppressor gene, p16INK4A. Deficient levels of MTAP can occur in cancers primarily through co-deletion of the MTAP gene and the p16INK4A gene. This antibody recognizes a protein of about 50 kDa, which is identified as gamma-enolase. Three isoenzymes of enolases are identified, alpha, beta and gamma. Alpha-isoform is expressed in most tissues, whereas beta-form is expressed predominantly in muscle tissue whereas gamma-enolase is found only in nervous tissue. These isoforms exist as both homodimers and heterodimers, and they play a role in converting phosphoglyceric acid to phosphenolpyruvic acid in the glycolytic pathway.
